ISTRUZIONI PER L'ACCESSO AL SERVIZIO DI "MUTATION DETECTION"

Va preparata secondo lo schema seguente:

E' importantissimo compilare la lista dei campioni scaricando il file Excel allegato (sia per i tubi sia per le micropiastre), in quanto il software consente l'introduzione diretta della lista evitando errori di battitura.
Le PCR devono essere effettuate in tubi da 0.2ml o micropiastre da 96 o 384.
Il volume minimo di PCR è 50 microlitri per il controllo e 25 microlitri per i campioni.
Le reazioni possono essere effettuate con la TaqGold (AB) o prodotto equivalente ultilizzando il buffer10x presente nella confezione della Taq Gold oppure il buffer LB 10x (TRIS base 100mM, HEPES acido 100mM, KCl 100mM, Solfato di ammonio 100mM, Cloruro di magnesio 15mM) senza olio minerale. Ogni altra possibile aggiunta (detergenti, DMSO, BSA, gelatina etc.) alla PCR è da evitare.
Se il controllo non è visibile o è contaminato da prodotti aspecifici nessun campione della stessa serie sarà caricato. Il controllo si può anche far pervenire con qualche gorno di anticipo oppure "una tantum", a patto che nel frattempo non si cambi il protocollo.
